Diversity of oligopeptide transporters in industrial yeasts

In winemaking fermentation nitrogen is a key nutrient, affecting both fermentation kinetics and the formation of wine aroma. The main nitrogen source for yeast are the ammonium and amino acids from the grape juice; nonetheless, oligopeptides have been reported to play a significant role in yeast metabolism and, more precisely, development of organoleptic compounds. Fungal Oligopeptide Transporters (FOT) are a novel family of oligopeptide transporters found in Saccharomyces wine strains. Recent studies have shown how Fot and other family transporters affect yeast nitrogen metabolism; however, little is known about the transport mechanism and its regulation. The aim of this project is to identify and characterize the oligopeptide transporters in Saccharomyces and non-conventional yeast commonly found in food ecosystems, as well as evaluate their impact on yeast fermentation performance. Genomic analysis, RT-PCR, phenotyping microarray or 14C analysis will be some of the techniques to perform in the analysis of transporters’ diversity and specificity. The effect of Fot transporters on fermentation, nitrogen consumption and flavour production will be studied using dedicated robotic and analytic platforms and assessed at pilot scale under industry conditions.
