Development of a model microbiological ecosystem of yeasts and bacteria to study the link between microbial diversity and resilience

In recent years, global warming, changes in agricultural practices, evolving consumer demands and the emergence of new technologies (high-speed sequencing, metagenomics) have led oenological research to focus on more integrative approaches that take into account microbial diversity and the ecological aspects. However, we lack relevant microbiological tools to answer these questions linking microbial diversity and the final quality of the oenological product.
The objective of this thesis is to design a model microbiological ecosystem representative of a grape must. Thanks to a methodological development combining molecular marking and fluorescence, we will be able to follow during an oenological fermentation the dynamic of frequencies of half-dozen species of yeast and bacteria that will compose it. This microbiological tool, which we consider indispensable today, will subsequently be made available to all players in the industry. In addition, we will use this microbiological model to answer critical questions on the impact of global warming (sugar, temperature) and the reduction of inputs (bioprotection, SO2) on the microbiological diversity of grape musts and its impact on wine quality. Thus, by varying its composition, we will be able to make the link between microbial diversity and ecosystem resilience.
